Next-Gen flow Cytometry & Sorting

OVERVIEW

Next-Gen flow Cytometry & Sorting (NGCS) facility at CAAD offers an efficient flow cytometric service to researchers by providing advanced support for training, experimental design, antibody panel development and data acquisition/analysis (software support). All-inclusive service is also supplied.

The service includes daily instrument calibration, sheat fluid, rinse fluid and clean fluid. Reagents and antibodies to mark the cells are in charge to each user.

The Unit is equipped with two cell analyzers: the BD FACSymphony™ A5 that leverages the inherent benefits of flow cytometry and enables the simultaneous measurement of up to 29 different characteristics of a single cell, the BD FACSCANTO II that offers automation and up to eight-color capacity for rapid and easy production of high-quality data for clinical and research purposes, and the BD FACSAria™ Fusion Flow Cytometer, that combines sorting capabilities (up to 14 markers) with best-in-class biosafety expertise for a comprehensive advanced cell sorter and biosafety solution.

These instruments are in line with the BD Rhapsody™ Single-Cell Analysis System that enables single-cell capture and barcoding of hundreds to thousands of single cells for analysis of genomic and proteomic information.

Recently, the Unit acquired: the Helios™ instrument, a CyTOF® system (Standard Bio tools) which serves as a high-dimensional single-cell suspension analysis tool and the Hyperion™ Imaging System which is an imaging mass cytometer that connected to the Helios instrument, is able to explore complex spatial relationship and bio-localize cells of interest within the tissue.

The potential of cytometers to detect simultaneously up to 30-50 parameters (physical and molecular) on a high number of cells per seconds (up to 10,000/40,000) make flow and mass cytometry the technologies of choice to count cells and simultaneously analyze several markers at the tissue levels, with high sensitivity in a very short period of time and with few amounts of biological samples or tissue. These approaches fit perfectly with basic research but also represent a powerful tool which can be used for diagnostic purposes and in clinical trials to assess immunotherapy candidates.

With regard to sorting, this is a restricted service that will be operated by facility staff only.

EQUIPMENTS

1) FACSymphony A5: It is an ultimate high-parameter flow cytometry system that allows the simultaneous measurement of up to 30 parameters. Specificities: 5 lasers (UV, violet, blue, yellow-green and red), 31 detectors (29 fluorescence + FSC and SSC) and FACSDiva 9.2 software (automatic stain index calculation).


2) FACSCanto II: It is an ease-of-use instrument that deliver reliable performance and accuracy and the High Throughput System (HTS) highly increase sample processing. Specificities: 3 lasers (violet, blue and red), 10 detectors (8 fluorescences + FSC and SSC), High throughput System (HTS).


3) FACSAria Fusion: It is a four-way sorter combined with best-in-class biosafety expertise for a comprehensive advanced cell sorter and biosafety solution. Specificities: 3 lasers (violet, blue and red), 16 detectors (14 fluorescneces + FSC and SSC), four-way sorting, plate sorting (48, 96, 384 etc), BSL2 biological hood and automatic drop delay calculation.

4) BD Rhapsody: Minimal set of benchtop equipment, very gentle cells manipulation (cells are settled into micro-well by gravity), highly stable magnetic barcoded beads, up to 40’000 cells per cartridge.

5) HELIOS: It is a high-dimensional single-cell mass cytometer able to detect metal-tagged antibodies (up to 45). Specificities: up to 135 channels and 350 antibodies/cell, replicate sample CV (normalized), automatic calibration. HYPERION: Scan area >1 mm2/2hr, frequency 200 pixel/sec, detection limit of > 400 copies per µm2, automatic calibration.
